On his father's side, McBride has ancestors from County Tyrone, Northern Ireland. His mother performed sermons in church using puppets, and McBride has stated that his "interest in telling stories comes from her". He was raised in Fredericksburg, Virginia, where he graduated from Courtland High School and attended North Carolina School of the Arts in Winston-Salem, North Carolina. McBride married Gia Ruiz in October 2010, at the Colony Palms Hotel in Palm Springs, California and had a son, Declan in 2011.

In 2006, McBride played Fred Simmons in the low-budget comedy film The Foot Fist Way, which he wrote with friends and frequent collaborators Jody Hill and Ben Best, with the Late Night with Conan O'Brien on February 26, 2008, and advertised the new soft drink flavor "mountain mist" for Gatorade. He appeared on comedy films Hot Rod, 30 Minutes or Less, Pineapple Express, Tropic Thunder, Observe and Report, Due Date, and Your Highness. He wrote and starred in the HBO comedy Eastbound & Down, about a washed up former major league baseball pitcher with anger management issues, also a collaboration with Hill and Best. The show was produced by Gary Sanchez Productions, the pilot episode premiered on February 15, 2009 and featured guest appearances from frequent collaborators Will Ferrell and Craig Robinson. In March 2009, McBride offered a contract to play a professional baseball with the Pensacola Pelicans, a minor league team in the American Association of Independent Professional Baseball, received the offer for his role as Kenny Powers in the show, and HBO announced it had renewed the series for a second season on April 8, 2009. In 2010, he signed an endorsement deal with K-Swiss shoes and HBO renewed Eastbound & Down for a fourth season on July 2, 2012. He played the eponymous roadie in the Tenacious D music video for Roadie in 2012.
